Definition Tylosin Tartrate is the tartrate of tylosin. The potency per mg should not be less than 800 Units, calculated with reference to the dried substance. Characteristics A white to pale yellow-coloured powder. Soluble in water, freely soluble in chloroform; slightly soluble in ethanol (96%); practically insoluble in ether. Identification A. The infrared absorption spectrum, Appendix II A, is concordant with the reference spectrum of Tylosin tartrate. B. The light absorption, Appendix II B, in the range 230 to 350 nm of a 0.004% w/v solution in 0.1M hydrochloric acid exhibits a maximum only at 290 nm. The absorbance at the maximum is about 0.88. C. To 10ml of a 0.004% w/v solution in 0.1M hydrochloric acid add 1 ml of 2 M sodium hydroxide, heat on a water bath for 20 minutes and cool. The light absorbance of the resulting solution, Appendix II B, in the range 250 to 430 nm, exhibits a maximum at 332 nm. D. Dissolve 30 mg in a mixture of 7.5 ml of pyridine, 2.5 ml of aceric anhydride and 0.15 ml of water and allow to stand. After about 10 minutes an emerald green colour is produced. Acidity or alkalinity pH of a 2.5% w/v solution, 5.0 to 7.2, Appendix VI. Tyramine Dissolve 50 mg in 5 ml of methanol in a 25 ml gradulated flask, add 2 ml of a 10% v/v solution of pyridine and 2 ml of a 2% w/v solution of ninhydrin. Close the flask by covering with a piece of aluminium foil and beat in a water bath at 85 of or exactly 30 minutes. Cool rapidly and add sufficient water to produce 25 ml. Use in the reference cell a solution prepared in a similar manner but omitting the substance being examined. The absorbance of the resulting solution, measured without delay, at 570 nm, Appendix II B, is not greater than that obtained by carrying out the procedure at the same time using 5 ml of a solution in methanol containing 35μg of tyramine per ml. Composition Carries out the method for liquid chromatograghy, Appendix III D, using solutions prepared immediately before useing in a mixture of equal volume of water and acetonotrile containing (1) 0.02% w/v of tylosin BPCRS and (2) 0.02% w/v of the substance being examined. The chromatographic procedure may be carried out using (a) a stainless steel column ( 20 cm long x 5 mm) packed with stationary phase C (5μm) ( Nucleosil C18 is suitable ), (b) as the mobile phase with a flow rate of 1 ml per minute, 0.85M sodium perchlorate in a 40% v/v solution of acetonurile the solution being adjusted to pH 2.5 using 1 M hydrochloric acid and (c) a detection wavelength of 290 nm. The chromatogram obtained with solution (1)shows similar resolution to the sample chromatogram supplied with the tylosin BPCRS. If necessary the molarity of the sodium perchlorate may be adjusted or the temperature of the column may be raised to a maximum of 50o. The order of elution of the six major components of tylosin BPCRS in the chromatogram obtained with solution (1) is desmycinosyltylosin, tylosin C, tylosin B, tylosin D, an aldol impurity and tylosin A. The column efficiency, determined using the peak due to tylosin A in solution (1), should be at least 22,000 theoretical plates per metre. Loss on drying when dried at 60°at a pressure not exceeding 0.7kl’a for 3 hours, loss not more than 5.0% of its weight. Use 1 g. sulphated ash not more than 3.0%, Appendix IX A. Use 1.67 g. Heavy metal not more than 20ppm, Appendix IX B. Assay Carries out a biological assay of antibiotics, Appendix XIV A. The precision of the assay is that the fiducial limit of error is not less than 95% and not more than 105% of the estimated potency. The upper fiducial limit of error is not less than 800 Units per mg, calculated with reference to the dried substance.
